Cal-520 AM provides a robust homogeneous fluorescence-based assay tool for detecting intracellular calcium mobilization. Cal-520 AM is a new fluorogenic calcium-sensitive dye with a significantly improved signal to noise ratio and intracellular retention compared to the existing green calcium indicators (such as Fluo-3 AM and Fluo-4 AM). Cells expressing a GPCR or calcium channel of interest that signals through calcium can be preloaded with Cal-520 AM which can cross cell membrane. Once inside the cell, the lipophilic blocking groups of Cal-520™ AM are cleaved by esterases, resulting in a negatively charged fluorescent dye that stays inside cells. Its fluorescence is greatly enhanced upon binding to calcium. When cells stimulated with agonists, the receptor signals the release of intracellular calcium, which significantly increase the fluorescence of Cal-520.

This detection kit is a robust, sensitive and easy-to-use assay to determine GPCR activity through detection of intracellular cAMP levels. The HitHunter cAMP assay is intended for use with cells expressing Gαs- or Gαi-coupled GPCRs. This product uses EFC technology, and contains ED-tagged cAMP, anti-cAMP antibody, and pure EA. Upon application to a cell which contains endogenous, untagged cAMP, the anti-cAMP antibody will be sequestered by binding to this untagged cAMP, allowing for EFC between the tagged cAMP and soluble EA. This will produce a proportional level of chemiluminescent signal. This kit format is intended for Small Molecules, with a protocol written for use in a 384-well plate. Mitochondria plays a crucial role in cells such as production of energy for cell viability. Mitophagy was recently found to be related to Alzheimer and Parkinson disease induced by the accumulation of depolarized mitochondria. Mitophagy serves as a specific elimination system that dysfunctional mitochondria caused by oxidative stress and DNA damage are sequestered into autophagosome, fused to lysosome and degraded by digestion. This kit is composed of Mtphagy Dye (reagent for detection of mitophagy) and Lyso Dye. Mtphagy Dye accumulates in intact mitochondria, is immobilized on it with chemical bonds and exhibits a weak fluorescence from the influence of surrounding conditions. When Mitophagy is induced, the damaged mitochondria fuses to lysosomes and then Mtphagy Dye emits a high fluorescence. To confirm the fusion of Mtphagy Dye labeled mitochondria and lysosomes, Lyso Dye included in this kit can be used.
